The move represents another setback for Mr Cameron involving the speech, which has been expected for months.
Sir Graham swiftly deleted the tweet, and later issued an apology, writing: "I wish to apologise profusely for the insensitive tweet I issued earlier. I have taken it down."
A Liberal Democrat spokesman said the comment was "wholly inappropriate and offensive".
"It's clearly sick and offensive and is utterly condemned by Nick Clegg," the spokesman told the BBC.
Sir Graham, the South West England and Gibraltar Liberal Democrat MEP, was knighted in the Queen's Birthday Honours in October 2011.
He said his knighthood was in recognition for his work introducing the controversial European Arrest Warrant and his leadership of the pro-EU Liberal Democrats in the European Parliament.
?We do not want to dignify this with a response. It speaks for itself,? said a spokesman for European Conservative MEPs.
